Editor’s Note: Over the past 20 years, Matt McCall has written up 50 stocks before they soared tenfold or more. Today he’s giving away a favorite new pick, free… at the center of what Dr. Mark Skousen calls “the most dangerous boom in history.” Click here to get the full details or read more below.
Dear Reader,
I recently sat down with Dr. Mark Skousen…
And he told me something that contradicts just about EVERYTHING we’ve been hearing about the stock market.
In short…
He says an astounding amount of money could flood into the markets in the years ahead.
We’re talking $60 trillion in new wealth up for grabs between now and 2029.
In my career, I’ve found 50 stocks before they soared tenfold or more.
(Most investors are lucky to see one ten-bagger in their lifetime.)
And I can tell you flat out…
I’ve never been more excited for what’s coming than I am right now.
See why here!
And as a special thank you for watching, you’ll get one of my favorite picks.
Regards,
Matt McCall

Alnylam’s Huntington’s Drug Cut the Disease Protein 64%. What the Model Is Really Pricing.
Analyst Targets
- Oppenheimer: Flagged a multibillion-dollar opportunity in Huntington’s disease following Friday’s data; called brezosiran a potential “best-in-class” knockdown agent
- UBS: Buy, $354 target (as of October 14)
- Citigroup: Buy, $320 target (as of October 14)
- BofA: Buy, $336 target (as of October 9)
- Wells Fargo: Equal Weight, $253 target
The Number That Changed the Conversation
In the ongoing Phase 1b study, participants on single ascending intrathecal doses of brezosiran showed robust, dose-dependent reductions of mutant huntingtin protein in cerebrospinal fluid. Across the first five cohorts, the highest dose group achieved a mean reduction of 60 to 64% at month three, with a maximum individual reduction of 83%. Those are not marginal numbers. For a disease where the protein is the disease, cutting it by more than half in spinal fluid with a single dose is a result the field has chased for years.
The company said the durability of the reductions suggests potential for twice-yearly dosing. Among participants followed for up to 12 months, no dose-dependent increases were observed in cerebrospinal fluid inflammatory markers, neurofilament light chain, or brain ventricular volume. That last point matters as much as the efficacy signal. Inflammation and neurofilament elevation were the warning signs that eventually derailed Roche’s tominersen. Brezosiran, at least through month 12, has shown neither.
Company Profile
Alnylam Pharmaceuticals is a leading biotechnology company specializing in RNAi therapeutics, headquartered in Cambridge, Massachusetts, with a market capitalization of approximately $30 billion. Its commercial engine runs on AMVUTTRA and other approved RNAi drugs in rare disease. Brezosiran, developed in a partnership with Regeneron signed in 2019 and anchored by $800 million in upfront cash and an equity investment, is the pipeline asset now generating the most clinical interest.
What the Data Show
- Mean CSF mutant huntingtin reduction: 60 to 64% at month 3 (highest dose cohort)
- Maximum individual reduction: 83%
- Study enrollment: 48 participants across five single-dose cohorts; a sixth cohort of 12 participants is currently enrolling
- Follow-up: up to 12 months, with no inflammatory signal detected
- Dosing potential: twice yearly, based on observed durability
The drug targets exon 1 of the huntingtin mRNA transcript, a design intended to reduce both full-length mutant HTT and the shorter mutant HTT1a. That dual knockdown is a differentiator. HTT1a has been identified as a driver of toxicity that some competing approaches leave untouched.
Why This Moves the Model
Huntington’s disease, a hereditary condition caused by mutant huntingtin protein accumulation, currently lacks effective disease-modifying treatments. That absence defines the commercial ceiling. If the therapy is ultimately approved, Alnylam could find itself with a multibillion-dollar opportunity in Huntington’s disease. Oppenheimer wrote Friday that “the severity of HD, coupled with lack of treatments, confers a significant market opportunity.”
The stock entered Friday near $227. Oppenheimer said the results fell in its “best case scenario,” suggesting “best-in-class knockdown.” That framing is significant: the firm was not surprised in an unexpected direction. The data cleared the internal bar analysts had modeled, which means target upgrades are more likely than initiation-level revisions.
Macro and Competitive Context
The Huntington’s field has a credibility problem, and brezosiran inherits it. Roche’s tominersen previously failed in Huntington’s, leaving the field without a validated RNAi or ASO readout until now. uniQure has advanced its gene therapy AMT-130 in clinical testing, while Wave Life Sciences continues to advance WVE-003, its allele-selective ASO, and PTC Therapeutics holds PTC518, an oral splicing modifier, in the pipeline. None have produced a Phase 1b RNAi knockdown of the magnitude Alnylam reported Friday.
The competitive read is straightforward: depth of protein knockdown is where brezosiran leads. Whether that biochemical lead converts to clinical benefit is the $2 billion question.
Forward Scenarios
Bull: The pivotal placebo-controlled trial, planned for 2027 pending regulatory feedback, shows statistically significant slowing of HD progression. With no approved disease modifier on the market, brezosiran gains accelerated approval and pricing power in a concentrated rare disease population. Analysts with $350-plus targets get covered.
Base: The trial enrolls on schedule and interim biomarker data confirm continued mHTT suppression. The stock re-rates modestly on trial initiation, then trades on timeline execution. Commercial upside is recognized only on approval, keeping the stock range-bound near current levels until 2028 data.
Bear: Protein knockdown does not translate to functional improvement, mirroring the trajectory of tominersen. Regulatory alignment takes longer than guided, and competitors close the gap. The pipeline premium compresses and the stock gives back gains driven by Friday’s data.
Technical Overlay
ALNY has traded in a compressed range since pulling back from its yearly highs, with resistance around $263 and $279. The stock was near $227 ahead of the brezosiran release. A sustained move above $263 on volume following Monday’s RNAi Roundtable, scheduled for October 26, would confirm that institutional buyers are treating this as a durable re-rating rather than a one-day catalyst. The SMA structure remains bearish on intermediate timeframes, so the burden of proof is on the upside.
What Investors Should Watch
- October 26 RNAi Roundtable: Brezosiran and the Phase 1b results will be featured as part of Alnylam’s 10th RNAi Roundtable series at 10 a.m. ET. Management tone on the pivotal trial timeline will drive the near-term move
- Q3 earnings, October 29: The company is scheduled to report results on October 29, 2026, before the market opens, with a conference call at 8:30 a.m. ET. Street revenue estimates cluster around $1.43 billion. AMVUTTRA trajectory remains the core commercial read
- Regulatory alignment confirmation for the 2027 pivotal study
- Sixth cohort data from the ongoing Phase 1b, which could extend the durability picture
- Analyst target revisions following Monday’s roundtable
Bottom Line
The brezosiran data are among the strongest early RNAi knockdown results in Huntington’s disease on record. Sixty to 64% mean knockdown, an 83% individual ceiling, and a clean 12-month safety readout: every number landed where bulls needed it. What the data do not yet show is functional benefit, and that is what the next trial is designed to establish.
The market is not yet paying for approval. At around $227, with targets ranging from $320 to $354 and investors still watching core execution in the commercial portfolio, ALNY trades well below where it would if brezosiran were viewed as a near-term revenue event. Friday’s data buy time and credibility. The pivotal trial converts one into the other, or it does not.
